In an interview with Games Radar, Splatoon producer Hisashi Nogami strongly hinted at DLC support for Nintendo’s summer squid title.

We can’t go into a lot of detail on that today, but we do have some plans to follow up with content to keep interest in the game post-launch. We on the development team are thinking of the launch as a first step of sorts. We hope to add to that in terms of content, and even to the degree that we’re hoping that this will become a franchise that Nintendo can be proud of.

We still have over a month before Splatoon will be released worldwide, so I doubt we will hear much more on the subject until this year’s E3 at the earliest.  The DLC could be new maps, modes, or even just new gear for your character in multiplayer.  Whatever the case, Nintendo has a decent track record with post game DLC thus far, so I have faith in this new team that we will see something worthwhile.
